Klimaet er meget koldt. Området har lange og kolde vintre, og korte kølige somre. Bjergene er dækket af sne hele året rundt. Årsnedbøren er på ca. 130 mm. Klimaet medfører at plantevæksten oppe på højsletterne er sparsomt, hovedsageligt græs med kun få træer, og nogle steder er der helt bart.

Der bor nogen hundrede tusind mennesker i Pamirområdet, hovedsageligt i dalene mod vest. Befolkningen består i vest af [[tadsjiker]] og på højsletterne i øst af nomadiske [[kirgisere]]. De vigtigste byer i området er [[Khorog]] i Pandzhdalen, [[Tashkurgan (by)|Tashkurgan]] i Xinjiang og [[Murgab]] i Gorno-Badakhsjan.
